Scientists Successfully Remove HIV From Human DNA for the first time



Once HIV invades the body, it doesn’t want to leave. Every strategy that scientists have developed or are developing so far to fight the virus – from powerful anti-HIV drugs to promising vaccines that target it – suffers from the same weakness. None can ferret out every last virus in the body, and HIV has a tendency to hide out, remaining inert for years, until it flares up again to cause disease.

None, that is, until now. Kamel Khalili, director of the Comprehensive NeuroAIDS Center at Temple University School of Medicine, and his colleagues took advantage of a new gene editing technique to splice the virus out of the cells they infected – essentially returning them to their pre-infection state. The strategy relies on detecting and binding HIV-related genetic material, and therefore represents the first anti-HIV platform that could find even the dormant virus sequestered in immune cells.

Even more encouraging, they also used the system to arm healthy cells from getting infected in the first place, by building genetic blockades that bounced off HIV’s genetic material. “It’s what we call a sterilizing cure,” says Khalili.

His work was done on human cells infected with HIV in cell culture, but, he believes the results are robust enough to move into animal trials and eventually into testing the idea in human patients.

The key to the strategy is the gene editing technique known as CRISPR, a way of precisely cutting DNA at pre-specified locations. CRISPR acts as a customizable pair of molecular scissors that can be programmed to find certain sequences of DNA and then, using an enzyme, make cuts at those locations. Because HIV is a retrovirus, its genetic material comes in the form of RNA; the virus co-opts a host cell’s genetic machinery to transform that RNA into DNA, which it then inserts into the cell’s genome. HIV’s genes, which it needs to survive, then get churned out by the cell.

Khalili designed a CRISPR that recognized the beginning and end of HIV’s DNA contribution, and then watched as the enzyme snipped out HIV from the cell’s genome. “I’ve been working with HIV almost since day 1 [of the epidemic] and we have developed a number of molecules that can suppress transcription or diminish replication of the virus. But I have never seen this level or eradication,” he says. “When you remove the viral genes from the chromosomes, basically you convert the cells to their pre-infection state.”

The advantage of the system lies in the fact that CRISPR can recognize viral genes wherever they are – in infected cells that are actively dividing, and in infected cells in which the virus is dormant. Current drug-based strategies can only target cells that are actively dividing and releasing more HIV, which is why they often lead to periods of undetectable virus but then cause levels of HIV to rise again. That’s the case with the Mississippi baby, who was born HIV positive and given powerful anti-HIV drugs hours after birth and appeared to be functionally cured of HIV when the virus couldn’t be detected for nearly four years, but then returned.

Khalili admits that more work needs to be done to validate the strategy, and ensure that it’s safe. But it’s the start, he says, of a potential strategy for eradicating the virus from infected individuals. That may involve excising the virus as well as bombarding it with anti-HIV drugs. “We can get into cells, eradicate the viral genome, and that’s it,” he says.

Weightlifter Chika Amalaha has won the first gold medal for Nigeria at the ongoing Commonwealth Games in Glasgow, United Kingdom, today Friday 25th July 2014. It is day 2 of the competition.
According to Wikipedia, Chika was born on the 28th of October 1997.
Amalaha pulled a total of 196 points in snatch and clean and jerk in the 53kg class. She beat weightlifters from Papa New Guinea Dika Toua and IndiaSantoshi Matsu, who won silver and bronze respectively.

Skales Dumps EME For Baseline Record Label

The rapper was unveiled by the label today Tuesday, July 22.

Skales who was with Banky W's EME but parted ways with the label after expiration of his contract in February 2014,will be joining female Pop singer Saeon in Baseline.

Although Skales is now an act under Baseline, he also announced the launch of his own record label, OHK Entertainment which he says will still run independently.

Lagos state grants six-month maternity leave, 10 days for fathers


Lagos state government has introduced a new leave regime for all male and female civil servants in the state. Henceforth, all male civil servants whose wives give birth are entitled to a 10-day paternity leave while female civil servants had their maternity leave extended from 3 months to 6 months.

The new leave regime was announced today by the Lagos State Head of Service, Mrs Josephine Williams at a press conference held at the Baguada Kaltho Press Centre, in Alausa, Ikeja.
"A female officer shall be entitled to 24-weeks maternity leave with full pay in case of her first two deliveries. The said leave shall commence, at least two weeks before the expected delivery date; a male officer, to who a new baby (or babies in case of multiple births) is born shall be entitled to 10 working days" she said.
She added that subsequent deliveries after the second baby by any employee will only get 12 weeks maternity leave for a female officer while the men will not be entitled to the 10-day leave.

Special adviser to the Lagos state Governor on public health, Dr Yewande Adeshina, said the 6 months leave will enable women complete the mandatory months required for breastfeeding new born babies.

Malaysia Airlines plane carrying 295 people crashes near Ukraine-Russia border

A Malaysia Airlines passenger plane carrying 295 people crashed near the Ukraine-Russia border Thursday, and a Ukraine official said later it had been shot down by a missile in the volatile region. Both government officials and pro-Russia separatists fighting in the region denied responsibility.
The number of fatalities in the crash was not immediately clear. An aide to Ukraine's interior minister quoted by Interfax said the total number of dead in the crash of Flight MH17 was more than 300 and includee 23 U.S. citizens. An Associated Press reporter at the scene reported seeing at least 22 bodies.
In brief comments made in Delaware Thursday afternoon, President Barack Obama called the crash a "terrible tragedy." He said the White House was working to determine whether there were U.S. citizens aboard the plane.
Meanwhile, the head of Ukrainian's emergency services told Reuters that search efforts at the scene of the crash we being hampered by "armed terrorists."
Anton Gerashenko, an adviser to Ukraine's interior minister, wrote on Facebook that the plane was flying at an altitude 33,000 feet when it was hit by a missile fired from a Buk launcher.
Separatist leader Andrei Purgin told the AP that he beieved Ukrainian troops had shot the plane down, but did not say why he was certain. Witnesses quoted by Russian media outlets said they saw a plane being hit by what they thought was a rocket, according to the news service.
Ukrainian President Petro Poroshenko said Ukraine's military was not responsible for the reported attack, which he called an “act of terror."
"We do not exclude that this plane was shot down, and we stress that the Armed Forces of Ukraine did not take action against any airborne targets," Poroshenko said. "We are sure that those who are guilty in this tragedy will be held responsible."
Malaysian Airlines earlier tweeted that it lost contact with Flight MH17 over Ukrainian airspace:

The airline later confirmed the crash, approximately 25 miles from the Russia-Ukraine border:
Flight MH17 operated on a Boeing 777 departed Amsterdam at 12.15pm (Amsterdam local time) and was estimated to arrive at Kuala Lumpur International Airport at 6.10 am (Malaysia local time) the next day. The flight was carrying 280 passengers and 15 crew onboard.

The plane crashed near a village called Hrabovo in the Donetsk region, an area that has seen clashes between Ukrainian forces and pro-Russia separatist rebels in recent days.
On Wednesday evening, a Ukrainian fighter jet was shot down by a Russian plane, Ukrainian authorities told the AP.
Prior to Thursday's crash, the U.S. Federal Aviation Administration, along with several other contries, had warned pilots not to fly over parts of Ukraine.
White House spokesman Josh Earnest said Obama directed his national security team to be "in close touch" with Ukrainian authorities. Obama spoke by phone with Russian President Vladimir Putin, at Moscow’s request, to discuss new sanctions announced by United States and European Union. According to the White House, Obama first learned of the crash during the talk with Putin.

This is the second tragedy to hit Malaysia Airlines in recent months. The Ukraine crash comes just four months after Malaysian Airlines Flight M370 lost contact less than an hour after takeoff from Kuala Lumpur International Airport and vanished. Adamawa state Governor Nyako impeached, deputy resigns position


Adamawa State House of Assembly members this afternoon July 15th impeached the state governor, Murtala Nyako, following a report by a 7-man investigative committee that probed allegations of gross misconduct and financial impropriety against him.

The report, which was read by the State Assembly Speaker Umaru Fintiri on the floor of the House, found the governor guilty of all the 16 allegations of gross misconduct. 18 out of the 24 members of the State Assembly then signed a resolution removing the governor.

The House then passed a motion that the Speaker, Umaru Fintiri, should be sworn in as the state’s acting governor. According to reports, that will be done by 4pm today.

Meanwhile, the state Deputy Governor who was also investigated by the panel, had resigned his position earlier in the day in a letter he sent to the House of Assembly which was read by members and immediately approved

“It might be our biggest game of the year,” Chief Executive Officer Niccolo de Masi said in an interview. “We’re not surprised. Kim is a one-of-a-kind talent with an incredibly precise fit to the game engine that we tailored but already had in the company.”
The Kardashian game takes users inside Hollywood, guided by a virtual Kim who offers advice on how to become an A-list celebrity, starting from the so-called E-list. “Dating famous people will get you more fans, too,” instructs Kardashian, who in real life is married to rap singer Kanye West.
While the game is free to play, the goal is to get users hooked on in-app purchases such as clothing or a burst of energy needed for traipsing through Hollywood. Users can spend as much as $99.99 for 175,000 virtual dollars. A trip to Beverly Hills costs 4 game “dollars,” while 400 will buy a necklace.
Kardashian’s voice guides the uninitiated through casting calls and parties, while users dodge surly tweets from rivals.
The game gives Kardashian’s most fervent fans a new way to satisfy their appetites, de Masi said.
“Whether you’re following Kim on Twitter, Facebook, Instagram or watching her show, with this game, you can play all the time,” de Masi said. “You can get your Kim interaction anytime you choose.”
Annual revenue from Kim Kardashian: Hollywood may reach $200 million, estimates Douglas Creutz, an analyst with Cowen & Co. in New York, who has the equivalent of a buy rating on Glu Mobile and expects the stock to rise to $6 in the next year.
It’s the only title among the top 10 in the App Store with a five-star rating, the highest possible, based on thousands of user reviews.
“There’s no such thing as bad publicity, right?” Creutz said. “Obviously, Kim Kardashian’s brand has driven people to download the game. But at this point, the game has taken on a life of its own.”

Lian Kao, a Taiwanese student recently became blind due to eyeball-eating amoebas.
Daily Mail reports that the hungry single-celled organisms formed after she knowingly left her contacts in for six months straight without changing or cleaning the lenses.
The lack of attention to eye hygiene created a breeding ground for an acanthamoeba infection to form in her contacts and her corneas. After the amoebas formed, they began burrowing into the student’s eyeballs, causing her to lose her vision.
She allegedly left the one pair of contacts in even while swimming and sleeping.
Wu Juan-liang, the director of ophthalmology at Taipei’s Wan Fang Hospital, explained.
“A shortage of oxygen can destroy the surface of the epithelial tissue, creating tiny wounds into which the bacteria can easily infect, spreading to the rest of the eye and providing a perfect breeding ground.
The girl should have thrown the contact lenses away after a month but instead she overused them and has now permanently damaged her corneas.” he said
